Broccoli Compound Targets Key Enzyme In Late-stage Cancer: "ScienceDaily (Dec. 2, 2008) — An anti-cancer compound found in broccoli and cabbage works by lowering the activity of an enzyme associated with rapidly advancing breast cancer...indole-3-carbinol, is already undergoing clinical trials in humans because it was found to stop the growth of breast and prostate cancer cells in mice..."I think one of the real uses of this compound and its derivatives is combining it with other kinds of therapies, such as tamoxifen for breast cancer and anti-androgens for prostate cancer," said coauthor Gary Firestone, UC Berkeley professor of molecular and cell biology. "Humans have co-evolved with cruciferous vegetables like broccoli and Brussels sprouts, so this natural source has a lot fewer side effects."...I3C inhibits the enzyme elastase, which at high levels in breast cancer cells heralds a poor prognosis: decreased response to chemotherapy, reduced response to endocrine treatment and reduced survival rates...Elastase is an enzyme that shortens a cellular chemical, cyclin E, that is involved in controlling the cell cycle. The shortened version of cyclin E accelerates the cell cycle, making cancer cells proliferate faster. Firestone showed that I3C prevents the elastase shortening of cyclin E, thereby arresting development of breast cancer cells...In previous work, they found that indole-3-carbinol interferes with more than cell proliferation. It also disrupts the migration and alters adhesion properties of cancer cells, as well as counteracts the survival ability of cancer cells, all of which are implicated in cancer cell growth. To have such broad downstream effects, I3C must act at the beginning of a major cellular pathway, Firestone said. The newly reported research pins this activity to elastase and its effect on cyclin E..."

Eating one or more portions of broccoli every week can reduce the risk of prostate cancer, and the risk of localised cancer becoming more aggressive...t was found that there were more changes in gene expression in men who were on the broccoli-rich diet than on the pea diet, and these changes may be associated with the reduction in the risk of developing cancer...Previous studies have suggested that the fifty percent of the population who have a GSTM1 gene gain more benefit from eating broccoli than those who lack this gene. The study showed that the presence of the GSTM1 gene had a profound effect on the changes in gene expression caused by eating broccoli...The results of the study suggested that relatively low amounts of cruciferous vegetables in the diet -- a few portions per week -- can have large effects on gene expression by changing cell signalling pathways. These signalling pathways are the routes by which information is transmitted through a molecular cascade which amplifies the signal to the nucleus of the cell where gene expression occurs...Cruciferous vegetables include broccoli, Brussels sprouts, cauliflower, cabbage, rocket, watercress, garden cress, kale, bok choy, radish, horseradish and wasabi.The broccoli used in this study is a high glucosinolate variety."

Lycopene no use in prostate cancer prevention

Study breaks link between lycopene and prostate cancer prevention EurekAlert 5/17/07 "...According to the researchers, the study is one of the largest to evaluate the role of blood concentrations of lycopene and other carotenoid antioxidants in preventing prostate cancer. Study data were derived from over 28,000 men enrolled in the Prostate, Lung, Colorectal, and Ovarian (PLCO) Cancer Screening Trial..."It is disappointing, since lycopene might have offered a simple and inexpensive way to lower prostate cancer risk for men concerned about this common disease," said Ulrike Peters, Ph.D., M.P.H..."

What a shame. The first research years ago seemed to indicate lycopene had good prevention qualities. Later studies were inconclusive. Now the largest and most comprehensive study seems to indicate lycopene is of no help.

But just as lycopene possibly fades away... up pops the benefits of cruciferous vegetables in preventing prostate cancer. Remember the term isothiocyanates (ITCs). I think we'll be hearing much more on these in the future. Phenethyl-ITC, or PEITC is the specific ITC related to preventing and fighting prostate cancer.

His laboratory has found that phenethyl-ITC, or PEITC, is highly effective in suppressing the growth of human prostate cancer cells at concentrations achievable through dietary intake..."Angiogenesis is a major issue in cancer metastases," said Dr. Singh. "Our results provide promising preliminary evidence that constituents of many edible cruciferous vegetables may slow down, or even halt, this process.""

Quite amazing that at dietary levels these vegetables can have a strong antiangiogenesis effect.
